  4. NAD+ works as fuel ( I guess NAD+ and ATP have similar functions, working for different departments, like gas and diesel.)
    With less NAD+, all the maintain pathway won’t work well.

  5. I guess , ATP provides fuel (energy) for protein, enzyme, membrane, moving and functioning.
    And NAD+ , provides fuel (energy) for mitochondrial, DNA transfer to RNA, telomerase, or DNA maintenance pathways, etc.

  8. Like you I think the interesting discussion is at points of disagreement. To provide some push back on the protein issue. First, I like that you mentioned many of the important factors here – mTOR, lean muscle mass.

    I have two criticisms. One, what you ended up saying explicitly has the ring of a trivial truth (ie uninformative and uncontested). It seemed to me that what you ended up saying explicitly was roughly, ‘don’t eat to little protein – you don’t want muscle wasting, but don’t eat too much – you don’t want overactivation of mTOR’. Sure, not too much, not too little, just right, we can all agree to that formula. That felt too vague.

    Second, the more substantive disagreement would be regarding what you’re implying – that the Goldilocks zone of protein intake does not involve any restriction of protein for most people, contrary to what Sinclair argues. Two of the issues you presented as needing a balance are activation of mTOR vs sarcopenia. Sinclair is drawn by the prospect of reducing mTOR activity, and you on the other hand talk about losing lean muscle mass and the importance of lean muscle. A few thoughts on weighing these factors. One, human protein requirements – what is necessary to avoid deficiency – is very low in comparison to Western intakes. Protein deficiency, known as kawshiorkor, is essentially non-existent in the west and mostly occurs in the context of starvation or malnutrition of the sort you see in famines. Since the issue here is not kwashiorkor, protein intake is a matter of which ‘biological program’ to favor, so to speak: growth or stress resistance and longevity. Outside of the old and frail (who are at high risk of sarcopenia largely due to inactivity, and falls secondary to sarcopenia), lower protein intake as Sinclair discusses does not pose risk for sarcopenia. A high protein diet is advised for the old inactive and frail specifically because they are uniquely at risk of sarcopenia, which is a risk factor for medical complications due to falls (hip fracture, hospitalization, etc). Someone who is younger and maintains an active lifestyle is not at risk for sarcopenia, and Maintaining notable lean muscle mass is compatible with lower protein intake. Of course, high protein intake can lead to more muscle mass… (As can taking insulin, IGF-1, other anabolic steroids etc). The point I’ve set up being that the trade off is between: (1) higher protein intake, more muscle growth and more muscle mass with the same amount of activity, and more mTOR activation, or (2) lower protein intake, less muscle growth (but no realistic risk of kawshiorkor or sarcopenia for the non-elderly and active), and less mTOR activation. Based on what I’ve read, it seems to me that the better course for long term health is lower protein intake compared to the norm, in the context of plenty of exercise. Another great researcher who discusses this topic quite a bit is Valter Longo from USC.

  13. Exercise and diet will ensure a healthy natural life but Sinclair’s results will help us to go beyond a natural life span. Our ancestors ate well and worked hard but they’re all dead. I want to live unnaturally long for a human. Like 300 years or more.

    • i was wondering the exact same thing!  This guy is, in my opinion, negating the PROOF of David Sinclair’s research, as well as his father’s and his own success in reversing aging by taking the supplements  David takes.  David and his team’s work with mice over the last several years using nmn and resveratrol are mindblowing to me….enough so that my husband and I are on the same protocol as he and his father.  This young 30 year old fella just wants to be heard.  What a shame, and we, in listening, and commenting, are giving him this much undeserved platform.  Oh well, lol, such is life and the surety that there will ALWAYS be someone to try and prove the successful wrong! There’s that 15 minutes of fame…..!
